My lab is interested in understanding cellular signalling processes in budding yeast using functional genomics tools.
First, we use synthetic genetic array (SGA) technology combined with a variety of libraries of yeast strains to genetically screen for interactions of enzymes (e.g. protein kinases, lysine deacetylases) and their targets. My group has created a variety of resources and collections for the community with application to the SGA platform, including a selection of overexpression arrays.
Second, we have combined SGA with high content screening (HCS) to assess changes in protein localization and abundance under a variety of genetic, environmental and chemical stresses using rapid image acquisition and analysis. SGA-HCS is also used to detect mutants that show defects in their sub-cellular morphology, such as reduced formation of DNA damage foci.
Third, my lab has pioneered the use of SGA for genome wide analysis of reporter gene-expression, particularly for studying transcriptional changes throughout the cell cycle. We are also interested in understanding cell cycle processes and molecular players that regulate endocytosis via directed mechanistic studies.
Finally, in collaboration with Dr. Charlie Boone, we are also trying to understand genetic redundancy and natural variation in yeast strains.

Most cells on earth spend the majority of their time in stationary phase. In yeast the transition from log phase to stationary phase has been associated with changes in abundance and localization of a number of proteins. I am interested in identifying changes in the proteome, using the GFP collection, with the goal of exploring how cells re-enter the cell cycle from stationary phase.

Ubiquitination is a major post-translational modification regulating a variety of key cellular processes. Deubiquitinases (DUBs) are therefore important modulators of cell physiology, attracting intense focus as therapeutic targets for molecular medicine. The role of deubiquitinases however remains largely enigmatic in yeast.
I am interested in systematic functional profiling of DUBs using a variety of SGA-based genetic screens and high-content imaging

A fitness defect from hemizygosity, or having only one functional allele of a gene, is known as haploinsufficiency, whereas complex haploinsufficiency (CHI) is the genetic phenomenon where hemizygosity at two or more loci in a diploid cell confers a fitness defect, indicating a genetic interaction. By combining automated yeast genetics with our temperature sensitive mutant collection of essential yeast genes, we plan to screen for all CHI interactions in the Saccharomyces cerevisiae essential yeast genome. Our hope is to discover novel genetic interactions and develop a CHI interaction network, which can partially explain the missing heritability problem.
